The anionic charged chromophores are repelled by the similarly negatively charged bacterial cells, resulting in transparent cells and a dark background. Acidic stains are more commonly named <b>negative stains<\/b>. Fewer negative stains exists. These are nigrosin stain, india ink, and Congo red.\r\n\r\nAn advantage of using negative staining is that heat fixation is not done, which allows more accurate determination of cell shape and size. It can also be used to see special structures, such as capsules, and is useful in observing spirochetes due to being very thin and small.\r\n\r\n&nbsp;\r\n\r\nMATERIALS\r\n\r\n<span style=\"text-decoration: underline\"><i>Each<\/i><i> <\/i><i>student<\/i><i> <\/i><i>should<\/i><i> <\/i><i>have<\/i>:\r\n<\/span>Blue rack\r\n2 glass slides\r\nNigrosin stain\r\nLens paper\r\nWindex (depends on instructor)\r\nInoculating loop\r\nBunsen Burner\r\nStriker\r\n1 culture of either <i>Escherichia<\/i><i> <\/i><i>coli<\/i><i> <\/i>or <i>Staphylococcus<\/i><i> <\/i><i>epidermidis\r\n<\/i>Microscope\r\n\r\n&nbsp;\r\n\r\nPROCEDURE OF NEGATIVE STAINING\r\n<ol>\r\n \t<li>Place a small drop of Nigrosin stain of the end of the slide.<\/li>\r\n \t<li>Using aseptic technique, transfer one loopful of bacteria into the drop of stain. Do not spread the stain and bacteria, but contain it as a drop at the end of the slide.<\/li>\r\n<\/ol>\r\n<div><img class=\"aligncenter\" src=\"https:\/\/press.wpunj.edu\/microbiologylabmanual\/wp-content\/uploads\/sites\/11\/2022\/11\/image1-7.jpeg\" alt=\"\" \/><\/div>\r\n3. Place the edge of the second slide (the spreading slide) into the drop of bacteria and stain until it spreads along the entire edge.\r\n<div><img class=\"aligncenter\" src=\"https:\/\/press.wpunj.edu\/microbiologylabmanual\/wp-content\/uploads\/sites\/11\/2022\/11\/image2-5.jpeg\" alt=\"\" \/><\/div>\r\n4. Push the spreading slide along the bottom slide, dragging the stain and bacteria evenly.\r\n<div><img class=\"aligncenter\" src=\"https:\/\/press.wpunj.edu\/microbiologylabmanual\/wp-content\/uploads\/sites\/11\/2022\/11\/image3-4.jpeg\" alt=\"\" \/><\/div>\r\n5.
